Cu-Ti-O納米管陣列的抗菌性能及其對內皮細胞和成骨細胞功能的影響

【摘要】:鈦及鈦合金具有優(yōu)良的生物相容性、良好的機械性能、耐腐蝕性及核磁共振成像的相容性等優(yōu)點,已被用作植入材料廣泛應用于整形外科領域,但是植入后的感染和松動等并發(fā)癥長期困擾醫(yī)患雙方。與傳統(tǒng)的微觀和宏觀形貌相比,納米級結構對調節(jié)細胞功能更加有效,其中通過陽極氧化在純鈦(Ti)表面構建的二氧化鈦(TiO_2)納米管陣列結構引起了人們的廣泛關注,因為適當尺寸的TiO_2納米管不僅可促進成骨細胞的成骨分化,也可促進內皮細胞的成血管能力。然而TiO_2納米管陣列抗菌能力差,無法有效對抗植入體感染。本研究首先在純Ti片表面利用磁控濺射技術沉積了不同Cu含量的TiCu涂層,然后通過陽極氧化成功地將Cu摻雜至TiO_2納米管陣列中,并研究了Cu摻雜TiO_2納米管陣列的長效抗菌性能、細胞相容性及其對內皮細胞和成骨細胞功能的影響。結果表明:(1)利用磁控濺射技術在純Ti表面沉積不同Cu含量的TiCu復合涂層,Cu的摻雜量可在0 at.%和20.04at.%之間調控;(2)當TiCu涂層中Cu的原子百分比不大于15.14%時,通過陽極氧化可以構建出高度有序的Cu-Ti-O納米管陣列;當TiCu涂層中Cu的原子百分比為20.04%時,制備出的為納米孔狀結構;(3)Cu-Ti-O納米管陣列持續(xù)釋放的銅離子(Cu~(2+))對革蘭氏陽性金黃色葡萄球菌表現(xiàn)出良好的長效抗菌能力;(4)所有納米管陣列表面對內皮細胞的粘附、活性影響不大,且適量的Cu摻雜能夠促進內皮細胞的鋪展,和TiO_2納米管陣列(對照組)相比,Cu-Ti-O納米管陣列表面及其離子提取物均能夠上調內皮細胞一氧化氮(NO)的合成和血管內皮生長因子(VEGF)的分泌,而且Cu-Ti-O納米管陣列的離子提取物培養(yǎng)內皮細胞的體外成血管能力也有所提高;(5)Cu-Ti-O納米管陣列對成骨細胞的堿性磷酸酶(ALP)活性無顯著影響;誘導分化早期,由于Cu~(2+)釋放量較大,Cu-Ti-O納米管陣列能夠促進Ⅰ型膠原蛋白的沉積,同時對細胞外基質礦化表現(xiàn)出一定的抑制作用,但隨著誘導培養(yǎng)時間的延長,這種現(xiàn)象變得越來越不明顯甚至消失。因此良好的抗菌性能、骨生成和血管生成能力使得Cu-Ti-O納米管陣列成為一種比較有前景的鈦植入體涂層。
[Abstract]:Titanium and titanium alloys have been widely used in plastic surgery due to their excellent biocompatibility, good mechanical properties, corrosion resistance and compatibility of nuclear magnetic resonance imaging. But complications such as infection and loosening after implantation have long plagued both doctors and patients. Compared with traditional microcosmic and macroscopic morphology, nanostructures are more effective in regulating cell function. Among them, TiO2 / TiO2) nanotube array structure constructed by anodic oxidation on the surface of pure titanium (Ti) has attracted much attention. The appropriate size of TiO_2 nanotubes can not only promote the osteogenic differentiation of osteoblasts, but also promote the vascular formation ability of endothelial cells. However, TiO_2 nanotube arrays have poor antibacterial ability and are not effective against implant infection. In this study, TiCu coatings with different Cu content were deposited on pure Ti substrates by magnetron sputtering technique, then Cu was successfully doped into TiO_2 nanotube arrays by anodic oxidation. The long-term antibacterial properties of Cu-doped TiO_2 nanotube arrays were studied. Cytocompatibility and its effects on endothelial cells and osteoblasts. The results show that the doping amount of Cu in TiCu composite coating with different Cu content deposited on pure Ti surface by magnetron sputtering technique can be adjusted between 0 at.% and 20. 04at.%) when the atomic percentage of Cu in TiCu coating is less than 15. 14? A highly ordered Cu-Ti-O nanotube array can be constructed by anodic oxidation, when the percentage of Cu atoms in the TiCu coating is 20.04km. The prepared nanostructured cupric copper ion (CuP2), which was continuously released from the nanoporous structure of Ti-Ti-O nanotube arrays, showed a good long-term antibacterial ability against Gram-positive Staphylococcus aureus.) all nanotube arrays adhered to endothelial cells on the surface of the nanotube arrays, and all nanotube arrays exhibited a long term antibacterial activity against S. aureus, and all nanotube arrays adhered to the endothelial cells on the surface of the nanotubes. The effect of Cu on the activity of endothelial cells was not significant, and Cu doping could promote the proliferation of endothelial cells. Compared with TiO_2 nanotube array (control group), the surface of Cu-Ti-O nanotube array and its ion extracts could up-regulate the synthesis of nitric oxide (no) and the secretion of vascular endothelial growth factor (VEGF) in endothelial cells. Moreover, the vascularization ability of endothelial cells cultured with ion extract of Cu-Ti-O nanotube array in vitro was also increased, and the activity of alkaline phosphatase (ALP) of osteoblasts was not significantly affected by Cu-Ti-O nanotube array, and early differentiation was induced. However, Cu-Ti-O nanotube array can promote the deposition of type I collagen protein and inhibit the mineralization of extracellular matrix, but with the prolongation of induction and culture time, Cu-Ti-O nanotube array can promote the deposition of type I collagen protein and inhibit the mineralization of extracellular matrix. This phenomenon becomes less and less obvious and even disappear. Therefore, good antibacterial properties, bone formation and angiogenesis make Cu-Ti-O nanotube arrays a promising coating for titanium implants.
