ABA調(diào)控?cái)M南芥種子休眠的相關(guān)機(jī)制探究

【摘要】:種子作為農(nóng)業(yè)生產(chǎn)中最基本、最重要的生產(chǎn)資料,植株的生長(zhǎng)發(fā)育和產(chǎn)量很大程度上取決于種子的質(zhì)量。種子的萌發(fā)和休眠在植物的個(gè)體生長(zhǎng)發(fā)育中處于初期階段,是一個(gè)至關(guān)重要的過(guò)程。探究種子休眠機(jī)制對(duì)提高種子活力,增加幼苗的抗逆性、出苗率以及作物產(chǎn)量都有著重大意義。植物激素脫落酸(ABA)能夠抑制種子的萌發(fā),促進(jìn)種子的休眠,其中ABI1基因具有非常重要的作用。ABI1基因在序列中第180處甘氨酸突變?yōu)樘於彼岷?導(dǎo)致種子對(duì)ABA不敏感。在野生型的擬南芥中非特異性表達(dá)該突變型ABI1基因,可消除種子對(duì)ABA的響應(yīng)。但這一影響是否存在組織特異性目前尚不明確。本研究通過(guò)構(gòu)建組織特異性啟動(dòng)子和植物GAL4/UAS異位表達(dá)技術(shù)在擬南芥的不同組織細(xì)胞中特異性表達(dá)abi1,利用不同濃度的ABA進(jìn)行萌發(fā)實(shí)驗(yàn),探究ABA在種子休眠的分子機(jī)制。研究結(jié)果如下:(1)根尖干細(xì)胞特異表達(dá)的abi1基因的轉(zhuǎn)基因植株種子萌發(fā)率高于野生型但萌發(fā)速率沒(méi)有明顯的增加。(2)莖尖干細(xì)胞特異表達(dá)的abi1基因的轉(zhuǎn)基因植株表現(xiàn)出對(duì)ABA更加敏感,萌發(fā)率不到40%。(3)J2133驅(qū)動(dòng)的abi1表達(dá)導(dǎo)致種子對(duì)ABA的敏感性降低。
[Abstract]:As the most basic and important means of production in agricultural production, the growth, development and yield of plants depend to a great extent on the quality of seeds.Seed germination and dormancy is a very important process in the process of individual growth and development of plants.It is of great significance to explore the mechanism of seed dormancy to improve seed vigor, increase seedling resistance, seedling emergence rate and crop yield.The plant hormone abscisic acid (Aba) can inhibit seed germination and promote seed dormancy. ABI1 gene plays a very important role. ABI1 gene mutates to aspartic acid at 180th point in the sequence, which results in the seed being insensitive to ABA.The nonspecific expression of the mutant ABI1 gene in wild type Arabidopsis thaliana could eliminate the response of seeds to ABA.However, it is not clear whether this effect has tissue specificity.In this study, we constructed tissue specific promoter and plant GAL4/UAS heterotopic expression techniques to express abi1 in different Arabidopsis tissues and cells, and used different concentrations of ABA to carry out germination experiments to explore the molecular mechanism of ABA in seed dormancy.The results are as follows: (1) the seed germination rate of abi1 gene specifically expressed by root tip stem cells is higher than that of wild type, but the germination rate of abi1 gene expressed by stem tip stem cells is not significantly increased. 2) the expression of transgenic plants of abi1 gene specifically expressed by stem tip stem cells is higher than that of wild type.More sensitive to ABA,Abi1 expression, which was less than 40%.(3)J2133 driven by germination rate, reduced the sensitivity of seeds to ABA.
