【摘要】：目的探討芒果苷(mangiferin,MA)對高果糖引起的自發(fā)性高血壓大鼠(spontaneously hypertensive rats,SHRs)腎間質纖維沉積的影響及其可能的機制。方法 24只SHRs按隨機單位組設計分組法分為對照組、果糖組、果糖+低劑量芒果苷(5 mg/kg)組和果糖+高劑量芒果苷(15 mg/kg)組。實驗周期為7周,7周末處死大鼠,取血漿、腎臟組織。檢測血漿葡萄糖、甘油三酯(TG)、總膽固醇(TC)、游離脂肪酸(non-esterified fatty acid,NEFA)和腎組織甘油三酯含量,Masson染色后半定量評估腎間質纖維沉積的程度,Real-time PCR檢測腎組織單核細胞趨化蛋白-1(monocyte chemoattractant protein-1,MCP-1)、CD68、α-平滑肌肌動蛋白(α-smooth muscle actin,α-SMA)、Ⅳ型膠原蛋白α-Ⅳ鏈[collagen alpha-4(Ⅳ)chain,col4a4]、轉化生長因子-β(transforming growth factor-beta,TGF-β)、纖溶酶原激活物抑制劑-1(plasminogen activator inhibitor-1,PAI-1)、尿激酶纖溶酶原激活物(urokinase plasminogen activator,u PA)mRNA的表達,免疫組化檢測腎臟u PA、PAI-1蛋白表達水平。結果與對照組比較,果糖組SHRs腎間質膠原纖維沉積率顯著增加(P0.01),而芒果苷(15 mg/kg)明顯減輕果糖引起的SHRs腎間質膠原纖維沉積(P0.05)。各組腎臟甘油三酯含量沒有變化;腎組織MCP-1、CD68、α-SMA、col4a4、TGF-βmRNA也沒有明顯變化,但芒果苷(15 mg/kg)明顯逆轉果糖引起的u PA/PAI-1 mRNA比值的降低(P0.05),同時降低高果糖引起PAI-1蛋白表達水平升高。結論芒果苷能改善高果糖所致SHRs腎臟間質纖維沉積,其機制可能與調控腎組織PAI-1、u PA有關。
[Abstract]:Objective to investigate the effect of mangiferin (mangiferin,MA) on renal interstitial fiber deposition in spontaneously hypertensive rats (spontaneously hypertensive rats,SHRs) induced by high fructose and its possible mechanism. Methods Twenty-four SHRs were randomly divided into control group, fructose low dose mangiferin group (5 mg/kg) and fructose high dose mangiferin group (15 mg/kg).
