【摘要】：理想三維支架的構(gòu)建是組織工程化人工器官成功的前提條件。以聚乳酸為模型材料,二氯甲烷為溶劑,利用超臨界流體誘導(dǎo)相分離過程制備具有外部三維立體結(jié)構(gòu)和內(nèi)部三維互通的組織工程支架,考察了過程制備的可行性,分析了操作參數(shù)如操作壓力、溫度和溶液濃度對孔的形態(tài)、結(jié)構(gòu)和孔壁的影響。結(jié)果表明,利用超臨界流體誘導(dǎo)相分離過程可成功制備孔徑分布均勻、互通性好、孔隙率高的組織工程用三維多孔支架,且制備周期短,工藝簡單。隨操作壓力、溫度和溶液濃度的增加,支架孔徑均逐漸減小。但過程中溫度太低,壓力或初始溶液濃度太高均會導(dǎo)致聚合物富相內(nèi)聚乳酸的快速結(jié)晶析出從而形成由納米級聚乳酸顆粒粘結(jié)在一起的支架孔壁。本實驗范圍內(nèi)較優(yōu)的操作參數(shù)為:高壓釜壓力10 MPa,溫度45℃,溶液濃度20%(質(zhì)量分?jǐn)?shù))。
[Abstract]:The construction of ideal three-dimensional scaffold is a prerequisite for the success of tissue engineering artificial organs. Using polylactic acid as model material and dichloromethane as solvent, tissue engineering scaffolds with external three-dimensional structure and internal three-dimensional interworking were prepared by supercritical fluid-induced phase separation process. The effects of operating parameters such as operating pressure, temperature and solution concentration on the pore morphology, structure and pore wall were analyzed. The results show that three-dimensional porous scaffolds for tissue engineering with uniform pore size distribution, good interoperability and high porosity can be successfully prepared by using supercritical fluid-induced phase separation process, and the preparation period is short and the process is simple. With the increase of operating pressure, temperature and solution concentration, the pore diameter of the scaffold decreases gradually. However, when the temperature is too low, the pressure or the initial solution concentration is too high, the rapid crystallization of poly (lactic acid) in the rich phase of the polymer will lead to the formation of the scaffold pore wall bonded by nano-sized polylactic acid particles.
