【摘要】：目的探討視黃醇結(jié)合蛋白4(RBP4)的蛋白表達(dá)量及RBP4 m RNA表達(dá)量在膽囊結(jié)石合并2型糖尿病組和未合并糖尿病組的膽囊組織中的差異。方法首先應(yīng)用免疫組化檢測30例病例組與30例對照組膽囊組織上RBP4的表達(dá)情況,然后用Western blot技術(shù)檢測膽囊黏膜組織上RBP4的表達(dá)量,并與相關(guān)臨床資料結(jié)合進(jìn)行統(tǒng)計(jì)學(xué)分析。結(jié)果免疫組化檢測到RBP4在病例組膽囊組織中的表達(dá)強(qiáng)度明顯高于對照組(P0.001),女性表達(dá)強(qiáng)度明顯高于男性(P0.001),腹型肥胖者表達(dá)強(qiáng)度明顯高于非腹型肥胖者(P0.001);Western blot結(jié)果顯示:RBP4在病例組的膽囊黏膜組織上的蛋白相對表達(dá)量為(2.91±0.86),對照組為(2.17±0.73),差異有統(tǒng)計(jì)學(xué)意義(P0.001),病例組RBP4在膽囊黏膜組織中的蛋白表達(dá)量明顯高于對照組。結(jié)論通過免疫組化和Western blot實(shí)驗(yàn),在分子生物學(xué)的層面上證明了2型糖尿病組膽囊組織的RBP4表達(dá)水平高于對照組,同時女性RBP4的表達(dá)強(qiáng)度高于男性,腹型肥胖者RBP4的表達(dá)強(qiáng)度高于非腹型肥胖者。
[Abstract]:Objective to investigate the expression of retinol binding protein 4 (RBP4) and the expression of RBP4 m RNA in gallbladder tissues of type 2 diabetes patients with gallstone and those without diabetes mellitus. Methods the expression of RBP4 in gallbladder tissues of 30 cases and 30 cases of control group was detected by immunohistochemistry, then the expression of RBP4 in gallbladder mucosa was detected by Western blot technique. Results Immunohistochemical staining showed that the expression of RBP4 in gallbladder tissue of the case group was significantly higher than that of the control group (P 0.001), the expression intensity of female was significantly higher than that of male (P 0.001), and the expression intensity of abdominal obesity was significantly higher than that of non-abdominal obesity (P 0.001). Western blot results showed that the expression of RBP4 in gallbladder tissue was significantly higher than that in control group. The relative expression of RBP4 in gallbladder mucosa was 2.91 鹵0.86 in the case group and 2.17 鹵0.73 in the control group. The difference was statistically significant (P 0.001). The expression of RBP4 in the gallbladder mucosa in the case group was significantly higher than that in the control group. Conclusion on the level of molecular biology, the expression of RBP4 in gallbladder tissue of type 2 diabetes mellitus is higher than that of control group, and the expression of RBP4 in women is higher than that in men. The expression of RBP4 in abdominal obesity was higher than that in non-abdominal obesity.
