細胞因子信號轉(zhuǎn)導抑制分子-3在肝纖維化中的作用及研究進展

【摘要】:肝纖維化是繼發(fā)于各種慢性肝損傷之后組織修復過程中的代償反應,其發(fā)病實質(zhì)是肝內(nèi)細胞外基質(zhì)(extracellular matrix,ECM)合成與降解動態(tài)平衡發(fā)生紊亂,最終導致ECM過度沉積。肝纖維化的發(fā)生與發(fā)展受到多種細胞因子及細胞內(nèi)多種信號轉(zhuǎn)導通路網(wǎng)絡的調(diào)控,其中很多靶點在肝纖維化中的作用尚沒有完全定論。近年來越來越多的研究證實細胞因子信號轉(zhuǎn)導抑制分子-3(suppressor of cytokine signaling,SOCS-3)在肝纖維化的發(fā)生與發(fā)展過程中扮演著重要角色。SOCS-3可通過調(diào)控肝臟內(nèi)炎癥反應、細胞增殖活化、胰島素抵抗、瘦素抵抗等影響肝臟疾病的發(fā)展進程。許多學者認為SOCS-3可作為肝纖維化疾病診斷、預測預后的生物分子指標以及治療靶標。該文從SOCS-3系統(tǒng)概述、SOCS-3與肝纖維化關系、SOCS-3在防治肝纖維化中的作用等方面綜述了SOCS-3在防治肝纖維化中的作用及研究進展。
[Abstract]:Liver fibrosis is a compensatory reaction in the process of tissue repair after various chronic liver injury. The essence of hepatic fibrosis is the disorder of the dynamic balance between the synthesis and degradation of extracellular matrix (extracellular matrix,ECM), which leads to the excessive deposition of ECM. The occurrence and development of hepatic fibrosis are regulated by many cytokines and many signal transduction pathway networks in cells, and the role of many targets in liver fibrosis has not been fully concluded. In recent years, more and more studies have confirmed that cytokine signal transduction suppressor molecule-3 (suppressor of cytokine signaling,SOCS-3 plays an important role in the pathogenesis and development of hepatic fibrosis. SOCS-3 can regulate the inflammatory response in the liver. Cell proliferation, insulin resistance and leptin resistance affect the development of liver disease. Many scholars believe that SOCS-3 can be used as a biomolecular index to predict the prognosis of liver fibrosis and as a therapeutic target. This article reviews the role of SOCS-3 in the prevention and treatment of hepatic fibrosis from the aspects of SOCS-3 system, the relationship between SOCS-3 and hepatic fibrosis, and the role of SOCS-3 in the prevention and treatment of hepatic fibrosis.
