玉米ZmPP6C基因的克隆及其響應(yīng)光質(zhì)和脅迫處理的表達(dá)模式分析

【摘要】:絲氨酸/蘇氨酸蛋白磷酸酶6亞基(catalytic subunits of Ser/Thr protein phosphatase 6,PP6C)是PP6全酶的催化亞基。在模式植物擬南芥中的研究表明,PP6C參與生長(zhǎng)素極性運(yùn)輸、脫落酸信號(hào)轉(zhuǎn)導(dǎo)和光信號(hào)轉(zhuǎn)導(dǎo)途徑介導(dǎo)的開(kāi)花調(diào)控。為了明確玉米絲氨酸/蘇氨酸蛋白磷酸酶6亞基(ZmPP6C)的蛋白結(jié)構(gòu)特征與同源蛋白間的進(jìn)化關(guān)系,采用RT-PCR方法克隆了ZmPP6C的全長(zhǎng)基因。序列分析表明,ZmPP6C開(kāi)放閱讀框?yàn)?12個(gè)核苷酸,編碼303個(gè)氨基酸殘基,包含PP2A的催化亞基PP2Ac結(jié)構(gòu)域;系統(tǒng)進(jìn)化樹(shù)分析表明,PP6C蛋白在進(jìn)化上較為保守,并且與高粱的PP6C蛋白相似性更高。對(duì)玉米自交系B73的ZmPP6C基因進(jìn)行器官特異性表達(dá)分析表明,其表達(dá)量在成株期葉片中最高,是根中的7.9倍;ZmPP6C能夠響應(yīng)不同光質(zhì)處理,且受遠(yuǎn)紅光和紅光的影響較大;也能響應(yīng)長(zhǎng)日和短日處理,在長(zhǎng)日條件下的光照和黑暗階段各有一個(gè)明顯的表達(dá)高峰,在短日條件下的光照和黑暗階段分別有2個(gè)和3個(gè)表達(dá)峰值;同時(shí),ZmPP6C還響應(yīng)高滲透、鹽漬和淹水等脅迫處理,出現(xiàn)明顯的上調(diào)表達(dá)。結(jié)果表明,ZmPP6C在玉米光信號(hào)轉(zhuǎn)導(dǎo)、開(kāi)花誘導(dǎo)與脅迫應(yīng)答中發(fā)揮重要作用,其分子與生化機(jī)制值得進(jìn)一步探討。
[Abstract]:Serine / threonine protein phosphatase subunit 6, catalytic subunits of Ser/Thr protein phosphatase 6PP6C, is the catalytic subunit of PP6 holozyme. Studies in Arabidopsis thaliana, a model plant, have shown that PP6C is involved in polar transport of auxin. Abscisic acid signal transduction and light signal transduction pathway mediated flowering regulation. In order to clarify the relationship between the protein structure characteristics of maize serine / threonine protein phosphatase 6 subunit ZmPP6C and the homologous proteins, The full-length gene of ZmPP6C was cloned by RT-PCR. Sequence analysis showed that the open reading frame of ZmPP6C was 912 nucleotides, encoding 303 amino acid residues and containing the PP2Ac domain of the catalytic subunit of PP2A. Phylogenetic tree analysis showed that PP6C protein was more conserved in evolution and more similar to PP6C protein in sorghum. The organ-specific expression of ZmPP6C gene in maize inbred line B73 was the highest in adult leaves. ZmPP6C in root could respond to different light quality treatments, and was influenced by far red light and red light, and could also respond to long day and short day treatment. At the same time, ZmPP6C was significantly up-regulated under high osmotic, salinized and flooded stress. The results showed that ZmPP6C was transduction in maize light signal transduction. The molecular and biochemical mechanisms of flowering induction and stress response should be further explored.
