組胺對脊髓α-運動神經元興奮性的易化作用及其離子機制(英文)

脊髓α運動神經元直接支配骨骼肌活動,是軀體運動和行為控制的最后公路(final common path)。因此,運動神經元興奮性調控對于運動行為的執(zhí)行至關重要。事實上,脊髓運動神經元接受多種神經調質的傳入,尤其是單胺能神經調質。然而,組胺和下丘腦組胺能神經系統對脊髓運動神經元的調控作用及其下游離子機制至今仍不清楚。本研究采用大鼠脊髓腦片細胞內記錄方法,發(fā)現組胺H1和H2受體的激活均能增強脊髓α運動神經元的重復放電行為和興奮性。鉀通道的關閉和鈉-鈣交換體的激活共同參與了H1受體激活介導的脊髓運動神經元的興奮,而超極化激活的環(huán)核苷酸門控(hyperpolarizationactivated cyclic nucleotide-gated, HCN)通道的開放則負責H2受體激活介導的興奮性效應。以上結果提示,通過切換與組胺受體相耦聯的離子通道和交換體的功能狀態(tài),組胺可以有效地偏置脊髓α運動神經元的興奮性,而下丘腦組胺能神經傳入很可能以這種方式直接調控最終的運動輸出并主動調節(jié)脊髓運動反射和運動執(zhí)行。
