藥物影響斗雞攻擊行為及其調(diào)控基因的研究

【摘要】:目的與意義:通過(guò)“藥物影響斗雞攻擊行為及其調(diào)控基因的研究”分析吐魯番斗雞攻擊行為與其血漿中皮質(zhì)醇(CORT)等激素、5-羥色胺轉(zhuǎn)運(yùn)體(5-HTT)等基因表達(dá)量的關(guān)系,探明影響吐魯番斗雞攻擊行為的影響因素。不僅有助于吐魯番斗雞攻擊行為性狀的選育以及其遺傳資源的保護(hù)和開(kāi)發(fā)利用,而且對(duì)其他動(dòng)物(包括人類)攻擊行為與內(nèi)分泌系統(tǒng)和5-HTT等基因的相關(guān)性的深入研究在理論與實(shí)踐上都具有重要意義。內(nèi)容與方法:以吐魯番斗公雞為試驗(yàn)材料,采用攻擊行為評(píng)分法、酶聯(lián)免疫法和QPCR等方法,分組研究不同劑量人用抗暴力性精神分裂癥的利培酮、阿立哌唑、曲唑酮、舍曲林4種藥物對(duì)吐魯番斗雞攻擊行為表型、CORT、MT、T、TSH的含量以及5-HTT、COMT、TPH1、DTNBP1基因的表達(dá)量的影響。結(jié)果與分析:(1)結(jié)果顯示口服利培酮等四種藥物后吐魯番斗雞的攻擊行為均呈現(xiàn)不同程度的減弱。對(duì)利培酮、阿立哌唑、、舍曲林(A4、B4組)均是第1天與第14天差異顯著(P0.05),舍曲林(C4組)第1天與第11、14天差異顯著(P0.05);其他組間、組內(nèi)均差異不顯著(P0.05)。(2)不同劑量利培酮和阿立哌唑組吐魯番斗雞血漿中CORT含量組內(nèi)均呈下降趨勢(shì),且隨著藥物劑量的增加和用藥時(shí)間的增長(zhǎng)趨于顯著水平,曲唑酮各組呈上升趨勢(shì)但差異不顯著(P0.05)。利培酮、曲唑酮組MT含量增加,阿立哌唑、舍曲林組組內(nèi)MT含量下降,且差異均不顯著(P0.05)。四種藥物對(duì)T和TSH含量的影響均不顯著(P0.05)。(3)利培酮和阿立哌唑使5-HTT基因的表達(dá)呈下調(diào)趨勢(shì),5-HTT:利培酮組A1、B1組第1、2、5與11、14天5-HTT的表達(dá)量差異顯著(P0.05);C1組第1、2與8、11、14天差異顯著(P0.05);阿立哌唑組:A2組第2與14天差異顯著(P0.05),B2組第1、2、5與8、11、14天差異顯著(P0.05),C2組第1與5、8、11、14天差異顯著(P0.05)。TPH1:利培酮組A1組第1與11差異極顯著(P0.01),第1、2、5、8與14差異顯著(P0.05),第2、5、8與11差異顯著(P0.05);B1組第1、2、5、8與11、14天差異顯著(P0.05);C1組1與5、8、11、14差異顯著(P0.05),第2與5、8、11差異顯著(P0.05),第2與14天差異極顯著(P0.01),阿立哌唑組:A2組第1、2與14差異顯著(P0.05),B2組第1與14差異顯著(P0.05),C2組第2與14天差異顯著(P0.05)。舍曲林、曲唑酮組5-HTT表達(dá)量呈上升趨勢(shì),對(duì)其他三種基因無(wú)明顯影響,差異不顯著(P0.05)。結(jié)論:四種人用治療精神分裂癥的藥物均可不同程度減少吐魯番斗雞的攻擊行為,其中以利培酮的減弱作用較為顯著。隨著吐魯番斗雞攻擊行為的減弱,CORT含量減少,利培酮、阿立哌唑組5-HTT基因含量降低,TPH1基因含量增加,說(shuō)明CORT、5-HTT基因與斗雞攻擊行為呈正相關(guān)、TPH1基因與斗雞攻擊行為呈負(fù)相關(guān),因此,可將CORT的含量與5-HTT基因、TPH1基因的表達(dá)量作為吐魯番斗雞攻擊行為選育的一個(gè)參考指標(biāo),為吐魯番斗雞攻擊行為的激素與分子選育奠定基礎(chǔ)。
[Abstract]:Aim and significance: to analyze the relationship between the aggressive behavior of Turpan cockfighting and the expression of 5-hydroxytryptamine transporter (5-HTT) in plasma by "study on the effect of drugs on aggressive behavior and its regulatory gene". The factors that influence the aggressive behavior of Turpan cocks are proved, which are not only helpful to the breeding of aggressive behavior traits of Turpan cocks, but also to the protection and utilization of their genetic resources. Furthermore, it is of great significance in theory and practice to study the relationship between aggressive behavior of other animals (including humans) and endocrine system and 5-HTT genes. Risperidone, aripiprazole, trazodone in different doses of human anti-violent schizophrenia were studied by aggressive behavior score, enzyme-linked immunosorbent assay (Elisa) and QPCR. The effects of sertraline on the content of TSH and the expression of DTNBP1 gene of 5-HTTTh1 TPH1 gene in Turpan cocks after oral risperidone were analyzed. The results showed that the aggressive behavior of Turpan cocks after taking risperidone and other four drugs was obvious. Varying degrees of abatement. For risperidone, Aripiprazole, sertraline A _ 4 / B _ 4) were significantly different on day 1 and day 14 (P 0.05), sertraline C 4) on day 1 and day 1114 (P 0.05). There was no significant difference in plasma CORT content between different doses of risperidone and aripiprazole in Turfan cocks, and it tended to be significant with the increase of drug dosage and time. The content of MT in trazodone group increased, while that in aripiprazole group and sertraline group decreased. No significant difference was found in the expression of 5-HTT gene between risperidone and aripiprazole. The expression of 5-HTT gene was down-regulated by risperidone and aripiprazole. There were significant differences on day 1 and day 1 between group 1 and group 2 on day 1, P 0.05, and on day 1, 2, and on day 1, 2, and on day 1, 1, 1, 1, 1, 1 and 14, respectively. The difference between group 1 and group 1 was significant (P 0.05), and that between group 1 and group 1 was significantly higher than that of group B 2 on day 14 (P 0.05). There were significant differences between 1 and 14 groups on the 2nd and 14th days of P0.05C _ 2 group (P 0.05), sertraline, sertraline (P 0.05), and sertraline (P 0.05). The expression of 5-HTT in trazodone group showed an upward trend, but had no obvious effect on the other three genes, but the difference was not significant (P 0.05). Conclusion: four kinds of drugs for treatment of schizophrenia can reduce the aggressive behavior of Turpan chickens in different degrees. With the decrease of attack behavior of Turpan cocks, the content of risperidone and aripiprazole reduced the content of 5-HTT gene, and the content of TPH1 gene increased with the decrease of attack behavior of Turpan cocks, and the decrease of 5-HTT gene content in risperidone and aripiprazole groups. These results suggested that the expression of CORT and the expression of 5-HTT gene could be used as a reference index for the selection of aggressive behavior of Turpan cockfighting. To lay a foundation for hormone and molecular breeding of attack behavior of Turpan cock.
