【摘要】：為探討促乳素釋放激素受體(prolactin releasing peptide receptor,PrRP-R)基因在不同發(fā)育時(shí)期綿羊睪丸和不同組織(心臟、肝臟、脾臟、肺臟、腎臟、背最長(zhǎng)肌、股二頭肌、下丘腦、垂體、松果腺)中表達(dá)的差異,以0,2,5,12,24月齡的小尾寒羊?yàn)檠芯繉?duì)象,應(yīng)用熒光實(shí)時(shí)定量PCR(quantitative real-time PCR)和免疫組化(immunohistochemistry)技術(shù)分別分析了PrRP-R基因在組織中的差異表達(dá)與定位。結(jié)果顯示:PrRP-R基因在睪丸與心臟、肝臟、脾臟、肺臟、腎臟、背最長(zhǎng)肌、股二頭肌、下丘腦、垂體、松果腺之間的表達(dá)差異極顯著(P0.01),股二頭肌、垂體、松果腺之間表達(dá)差異不顯著(P0.05),但背最長(zhǎng)肌、下丘腦、垂體之間表達(dá)差異顯著(P0.05);0,2,5,12,24月齡的睪丸組織中均有一定量的表達(dá),24月齡時(shí)表達(dá)最高(P0.01);PrRP-R在0~24月齡綿羊睪丸組織中呈階段性表達(dá),0,2,5月齡時(shí),可觀察到PrRP-R在精原細(xì)胞中到陽(yáng)性表達(dá);12,24月齡時(shí),在初級(jí)精母細(xì)胞和次級(jí)精母細(xì)胞中檢測(cè)到強(qiáng)陽(yáng)性表達(dá)。結(jié)果表明:PrRP-R在不同的組織和睪丸中表達(dá)并存在一定的差異,可能對(duì)綿羊的繁殖性能具有重要的調(diào)控作用。
[Abstract]:To investigate the expression of prolactin releasing peptide receptor PrRP-RR gene in testis and tissues (heart, liver, spleen, lung, kidney, longissimus dorsi, biceps femoris, hypothalamus, pituitary) of sheep at different developmental stages. The difference of expression in pineal gland was studied in small tail Han sheep aged 24 months. The differential expression and localization of PrRP-R gene in tissues were analyzed by fluorescence real-time quantitative PCR(quantitative real-time and immunohistochemical histochemistry.The results showed that the PrRP-R gene was expressed in testis and heart, liver, spleen, lung, kidney, longissimus dorsi muscle and biceps femoris. The difference of expression among hypothalamus, pituitary and pineal gland was very significant (P 0.01). There was no significant difference in expression between biceps femoris, pituitary and pineal gland, but the longest dorsal muscle and hypothalamus. There was a significant difference in the expression of PrRP-R between the pituitary gland and the testis at 24 months of age. The highest expression of PrRP-R was found in the testis at 24 months of age. The expression of PrRP-R in the testis of 0 to 24 months old sheep showed a periodic expression at the age of 0 to 24 months, and the expression of PrRP-R in the testis of sheep at the age of 0 to 24 months was at the age of 0 to 2 months. Strong positive expression of PrRP-R was observed in primary spermatocytes and secondary spermatocytes at 24 months of age. The results showed that there were some differences in the expression of PrRP-R in different tissues and testis. It may play an important role in regulating the reproductive performance of sheep.
