解淀粉芽孢桿菌對(duì)免疫抑制肉雞生長性能和肝臟抗氧化功能的影響

【摘要】:旨在研究解淀粉芽孢桿菌(Bacillus amyloliquefaciens,BA)對(duì)環(huán)磷酰胺(cyclophosphamide,CY)介導(dǎo)的免疫抑制肉雞生長性能和肝臟抗氧化功能的影響。144只1日齡AA雄性肉仔雞被隨機(jī)分為3組,即對(duì)照組(CON),環(huán)磷酰胺處理組(CY)和環(huán)磷酰胺-解淀粉芽孢桿菌組(CY-BA),每組6個(gè)重復(fù),每個(gè)重復(fù)8只雞。CON組和CY組飼喂基礎(chǔ)日糧,CY-BA組飼喂添加1.08×1010 CFU/kg BA的試驗(yàn)日糧。于16,17和18日齡對(duì)CY組與CY-BA組肉雞肌肉注射80mg/kg的CY,CON組肌肉注射等量生理鹽水,試驗(yàn)期為21d。結(jié)果顯示,與CON組相比,CY刺激顯著降低了CY組肉雞1~21日齡體增質(zhì)量、肝臟指數(shù)、肝臟總抗氧化能力(total antioxidative capacity,T-AOC)、谷胱甘肽過氧化物酶(glutathione peroxidase,GSH-Px)活力和谷胱甘肽還原酶(glutathione reductase,GR)活力(P0.05),顯著升高了肝臟丙二醛(malondialdehyde,MDA)含量(P0.05);Real-time PCR檢測也發(fā)現(xiàn),CY處理顯著降低了CY組肉雞肝臟核轉(zhuǎn)錄相關(guān)因子2(NF-E2-related factor 2,nrf2)和谷胱甘肽過氧化物酶1(glutathione peroxidase 1,gpx1)的mRNA表達(dá)水平(P0.05)。與CY組相比,日糧添加BA顯著提高了CY-BA組肉雞21日齡時(shí)肝臟T-AOC水平和GSH-Px活力,降低了肝臟MDA水平(P0.05)。上述指標(biāo)在CON與CY-BA組間并無明顯差異(P0.05)。另外,CY-BA組肝臟GSH含量較CON或CY組均顯著升高(P0.05)。然而,日糧添加BA并未顯著影響CY刺激肉雞體增質(zhì)量、肝臟指數(shù)、肝臟nrf2和gpx1mRNA水平(P0.05)。綜合結(jié)果表明,CY誘導(dǎo)的免疫抑制損傷了肉雞生長和肝臟的抗氧化功能,并引起了肉雞肝臟脂質(zhì)過氧化程度升高。日糧添加BA具有改善CY介導(dǎo)的免疫抑制肉雞肝臟抗氧化功能和緩解肝臟氧化損傷的作用。
[Abstract]:The purpose of this study was to study the effects of Bacillus amyloliquefaciens,BA on the growth performance and liver antioxidant function of immunosuppressive broilers mediated by cyclophosphamide (cyclophosphamide,CY). 144 1-day-old AA male broilers were randomly divided into three groups: control group (CON), cyclophosphamide treatment group (CY) and cyclophosphamide-starch-hydrolyzed spores group (CY-BA), with 6 repeats in each group. The basal diet was fed in CON group and CY group, and the experimental diet supplemented with 1.08 脳 1010 CFU/kg BA was fed in CY-BA group. At the age of 16, 17 and 18 days, the broilers in CY group and CY-BA group were injected with 80mg/kg intramuscular injection of the same amount of normal saline, the experimental period was 21 days. The results showed that compared with CON group, CY stimulation significantly decreased the weight gain, liver index, total antioxidant capacity (total antioxidative capacity,T-AOC, glutathione peroxidase,GSH-Px activity and glutathione reductase,GR activity of broilers in CY group (P 0.05), and significantly increased the content of malondialdehyde (malondialdehyde,MDA) in liver (P 0.05). Real-time PCR assay also showed that CY treatment significantly decreased the mRNA expression of nuclear transcription related factor 2 (NF-E2-related factor 2, nrf2) and glutathione peroxide 1 (glutathione peroxidase 1, gpx1 in CY group (P 0.05), and decreased the expression of nuclear transcription related factor 2 (nrf2, nrf2) and glutathione peroxide 1 (Gpx1) in liver of broilers in CY group (P 0.05). Compared with CY group, dietary BA significantly increased T-AOC level and GSH-Px activity in liver and decreased liver MDA level in CY-BA group at 21 days of age (P 0.05). There was no significant difference between CON and CY-BA (P 0.05). In addition, the content of GSH in liver of CY-BA group was significantly higher than that of CON or CY group (P 0.05). However, dietary BA supplementation did not significantly affect body weight gain, liver index, liver nrf2 and gpx1mRNA levels in broilers stimulated by CY (P 0.05). The results showed that immunosuppression induced by CY damaged the growth of broilers and the antioxidant function of liver, and increased the degree of lipid peroxide in liver of broilers. Dietary addition of BA can improve the antioxidant function of liver mediated by CY and alleviate the oxidative damage of liver in broilers.
