【摘要】：目的檢測ADAM8基因在胃癌組織中的表達水平,探討胃癌的生物學特征與其表達之間的關系。方法應用免疫組織化學、實時定量PCR及Western blot技術檢測100例胃癌組織、20例正常胃黏膜組織中ADAM8基因的表達,并分析其與臨床病理特征的相關性。結果免疫組織化學分析顯示ADAM8在正常胃黏膜組織中表達率為30%(6/20),而胃癌組織中ADAM8的表達率為86%(86/100),提示ADAM8基因表達在胃癌組織中顯著上調(diào)(P0.05);實時定量PCR檢測顯示在胃癌標本中ADAM8 mRNA的表達水平為(0.767±0.129),在正常胃組織中表達水平為(0.192±0.127),兩者差異有統(tǒng)計學意義(P0.01)。Western blot檢測結果顯示在胃癌組織中ADAM8蛋白表達水平為(0.659±0.027),而正常胃黏膜標本中其表達水平為(0.236±0.074),差異有統(tǒng)計學意義(P0.01)。且伴淋巴結轉(zhuǎn)移胃癌組織、無淋巴結轉(zhuǎn)移胃癌組織與正常胃黏膜組織相比,ADAM8蛋白表達水平明顯增高,差異均具有統(tǒng)計學意義(均P0.05)。進一步分析ADAM8蛋白表達與臨床病理特征之間的關系,結果顯示其表達與腫瘤的直徑、臨床分期、腫瘤的浸潤深度、有無轉(zhuǎn)移等顯著相關(均P0.05);與性別、年齡等因素無明顯相關(均P0.05)。結論 ADAM8在胃癌組織中的高表達與胃癌侵襲轉(zhuǎn)移有關,提示ADAM8可能參與胃癌的發(fā)生、發(fā)展過程,可能成為胃癌預后判斷的參考指標。
[Abstract]:Objective to detect the expression level of ADAM8 gene in gastric carcinoma and to explore the relationship between its biological characteristics and its expression. Methods Immunohistochemistry was used. Real-time quantitative PCR and Western blot techniques were used to detect the expression of ADAM8 gene in 20 normal gastric mucosa tissues. Results Immunohistochemical analysis showed that the expression rate of ADAM8 in normal gastric mucosa was 30 / 6 / 20). The expression rate of ADAM8 in gastric cancer was 86 / 100, suggesting that the expression of ADAM8 gene was significantly up-regulated in gastric cancer tissues. The expression level of ADAM8 mRNA in gastric cancer was 0.767 鹵0.129 by real-time quantitative PCR. The expression level in normal gastric tissues was 0.192 鹵0.127). The expression level of ADAM8 protein in gastric carcinoma was 0.659 鹵0.027). The expression level in normal gastric mucosa was 0.236 鹵0.074, the difference was statistically significant (P 0.01). The expression of ADAM8 protein in gastric cancer without lymph node metastasis was significantly higher than that in normal gastric mucosa. The relationship between the expression of ADAM8 protein and the clinicopathological features was further analyzed. The results showed that the expression of ADAM8 protein was related to the diameter and clinical stage of the tumor. The depth of invasion and metastasis were significantly correlated (all P 0.05); Conclusion the high expression of ADAM8 in gastric cancer is related to invasion and metastasis of gastric cancer, suggesting that ADAM8 may be involved in the occurrence of gastric cancer. The process of development may be a reference index for judging the prognosis of gastric cancer.
