羊駝皮膚Scf基因的分子克

干細(xì)胞因子(SCF)是一種生長(zhǎng)因子,在黑素細(xì)胞遷移、細(xì)胞增殖和分化中發(fā)揮重要的調(diào)節(jié)作用。為了探討SCF的分子特性及其在不同毛色羊駝皮膚中的表達(dá)模式,本研究通過(guò)PCR克隆羊駝Scf基因的全長(zhǎng)編碼區(qū)(CDS),并運(yùn)用生物信息學(xué)預(yù)測(cè)其編碼蛋白質(zhì)特征;采用實(shí)時(shí)定量PCR (qRT-PCR)、Western印跡和免疫熒光分析SCF的表達(dá)模式和組織分布。結(jié)果表明,Scf編碼區(qū)核苷酸為740 bp,編碼1個(gè)246個(gè)氨基酸的前體蛋白質(zhì)。該蛋白質(zhì)包含1個(gè)跨膜區(qū)和1個(gè)信號(hào)肽,其二級(jí)結(jié)構(gòu)核心是4個(gè)α-螺旋(αA,αB,αC和αD)和2條反平行的β-折疊鏈;序列比對(duì)和系統(tǒng)發(fā)育分析顯示,羊駝SCF序列與豬、牛、羊同屬于一個(gè)分支;棕色羊駝皮膚SCF表達(dá)顯著高于白色羊駝,SCF蛋白主要位于毛囊基質(zhì)與毛根鞘中,且Fontana-Masson染色顯示,棕色羊駝皮膚中含有更多的黑色素顆粒。以上研究結(jié)果證實(shí),在棕色羊駝毛皮膚中SCF的表達(dá)量和黑色素含量均高于白色羊駝毛皮膚,且SCF的高級(jí)結(jié)構(gòu)與毛色形成密切相關(guān)。
