【摘要】:目的膠原是治療骨軟骨缺損的一種常用生物材料,然而目前膠原支架的結(jié)構(gòu)大多是無序的,因此不利于自體細(xì)胞向缺損部位遷移進(jìn)行缺損修復(fù)�;|(zhì)細(xì)胞衍生因子-1(SDF-1)被認(rèn)為能夠促進(jìn)間充質(zhì)干細(xì)胞向軟骨下骨遷移。本實驗?zāi)康氖峭ㄟ^設(shè)計有取向通道的膠原支架復(fù)合SDF-1來促進(jìn)細(xì)胞向缺損部位遷移,從而促進(jìn)骨軟骨缺損的修復(fù)和治療。方法我們首先通過膠原凍干和熱交聯(lián)制作了無序膠原支架和有序膠原支架,分別用電鏡對支架通道進(jìn)行表征,并對它們進(jìn)行了力學(xué)性能,細(xì)胞毒性,細(xì)胞增殖等檢測。然后我們檢測了骨髓間充質(zhì)干細(xì)胞在材料內(nèi)的遷移。最后,我們在新西蘭兔上建立了骨軟骨缺損模型,然后將兩種膠原支架分別復(fù)合SDF-1移植到缺損部位,分別在6周和12周收集樣本,通過ICRS評分、番紅-固綠染色以及免疫組化染色檢測軟骨再生情況。結(jié)果有序膠原支架較無序膠原支架有更好的力學(xué)強(qiáng)度,兩種材料對細(xì)胞增殖無明顯影響。有序膠原支架可以促進(jìn)骨髓間充質(zhì)干細(xì)胞的遷移,該作用能夠被SDF-1加強(qiáng)。動物實驗結(jié)果表明結(jié)合有序膠原支架復(fù)合細(xì)胞因子SDF-1可以促進(jìn)骨軟骨缺損部位新生軟骨的生長,同時能夠抑制軟骨細(xì)胞的終末分化和鈣化。結(jié)論通過有序膠原支架復(fù)合SDF-1的移植是一種可行的骨軟骨缺損療法。
[Abstract]:Objective Collagen is a commonly used biomaterial for the treatment of osteochondral defects, but the structure of collagen scaffold is disordered. This is not conducive to the migration of autologous cells to the defect site for repair. Stromal Cell derived Factor-1 SDF-1). It is believed that mesenchymal stem cells can promote the migration of mesenchymal stem cells to subchondral bone. In order to promote the repair and treatment of osteochondral defects. Methods We first prepared the disorder collagen scaffold and the ordered collagen scaffold through the collagen freeze-drying and thermal crosslinking, and used electron microscope to characterize the scaffold channel. Then we detected the migration of bone marrow mesenchymal stem cells in materials. Finally, we established a bone cartilage defect model in New Zealand rabbits. Then the two collagen scaffolds combined with SDF-1 were transplanted to the defect site, and the samples were collected at 6 and 12 weeks, respectively, and the ICRS score was used. Results the ordered collagen scaffold had better mechanical strength than the disordered collagen scaffold. The two materials have no significant effect on cell proliferation. Ordered collagen scaffolds can promote the migration of bone marrow mesenchymal stem cells. The effect could be enhanced by SDF-1. The results of animal experiments showed that the combination of ordered collagen scaffold and cytokine SDF-1 could promote the growth of new cartilage in bone cartilage defect. At the same time, it can inhibit the terminal differentiation and calcification of chondrocytes. Conclusion the transplantation of SDF-1 combined with ordered collagen scaffold is a feasible treatment for osteochondral defects.
