【摘要】：全能干細胞主要包括胚胎干細胞(embryonicstem cell,ESC)和自體誘導多能干細胞(induced pluripotent stem cell,iPSC)兩類,其在肝臟方向的分化及移植應用,可為肝細胞替代治療提供新的細胞來源,有望成為終末期肝病的有效輔助療法.近年干細胞技術在肝臟領域研究進展迅速,如iPSC可在保留全能分化性的同時大大減輕ESC應用面臨的倫理及免疫排斥問題,同時干細胞分化水平也逐漸從肝系細胞的單細胞狀態(tài)提升到肝臟組織的高度.這些進展一方面推動了肝功能替代治療技術進步;另一方面也促進了肝臟器官體外發(fā)育基礎理論的豐富和更新.
[Abstract]:Totipotent stem cells mainly include embryonic stem cell ESCs and autologous induced pluripotent stem cells induced pluripotent stem cell sci SC.Their application in liver differentiation and transplantation can provide a new cell source for hepatocyte replacement therapy. It is expected to be an effective adjuvant therapy for end-stage liver disease. In recent years, stem cell technology has made rapid progress in the field of liver research. For example, iPSC can significantly alleviate the ethical and immune rejection problems in the application of ESC while preserving its totipotent differentiation. At the same time, the differentiation level of stem cells gradually increased from the single cell state of liver cells to the high level of liver tissue. These advances not only promote the progress of liver function replacement therapy, but also promote the enrichment and renewal of the basic theory of liver organ development in vitro.
